Numerous new synthesis methodologies are developped which can be applied to heterocycles and more particularely to purines. These methodologies include regioselective functionalizations of the purine nucleus by metalation, Suzuki and Sonogashira cross-couplings, or by building purine derivatives from pyrimidine precursors. The synthesis of protein kinase targeted libraries has been the subject of many works which have led to the identification of several protein kinase inhibitors (CDK1, CDK2, CDK5, GSK3-b, CK1, Tyro3, Met).
